Dual Inhibition of IDO1 and TDO: A unified therapeutic strategy to combat Alzheimer's Diseases and Cancer

In this study, we explored a dual-targeting approach against the kynurenine pathway enzymes Indoleamine 2,3-dioxygenase 1 (IDO1) and Tryptophan 2,3-dioxygenase (TDO)revealing their synergistic inhibition as a promising strategy for tackling both neurodegeneration and tumor progression.
The study offers valuable insights into:

  • The structural and energetic basis of dual inhibition.

  • Lead-like phytochemicals that show strong binding and specificity toward both targets. A common mechanistic strategy that connects immune evasion in cancer and neuroinflammation in Alzheimer’s disease, offering a unified therapeutic avenue.
